Factors responsible for the abnormal development of embryos obtained by nuclear transplantation in Xenopus laevis.

چکیده
IN Xenopus the embryos derived from nuclear transplantation often develop abnormally. These abnormalities must be due to the limited potentiality for development of either the donor nucleus or the egg cytoplasm; this limited potentiality will in turn be due to technical damage during transplantation or to the innate condition of the nucleus or cytoplasm before the experiment. The extent to which these technical and innate factors are responsible for abnormalities of transplant-embryo development has been analysed by considering the effect of each factor in turn. Nuclei from early donor stages have been used, since these nuclei are believed to be undifferentiated (see p. 338) and therefore to have the innate capacity for entirely normal development. The effects of other factors have been investigated by experiments in which each factor is varied in different ways. Any correlation between variations in one factor and the resulting proportion of abnormal transplant-embryos is then recorded. In this way the effect of each factor on the development of transplant-embryos can be analysed. These experiments show how nuclei can be transplanted with least damage, and whether small inconsistencies in carrying out the technique can bring about abnormalities of development. It is thus made clear to what extent abnormal transplant-embryos may be considered to represent innate qualities of donor nuclei.

Expression of hsp90 Alpha and hsp90 Beta during Xenopus laevis Embryonic Development
Background: Members of the eukaryotic Hsp90 family function as important molecular chaperones in the assembly, folding and activation of cellular signaling in development. Two hsp90 genes, hsp90 alpha and hsp90 beta, have been identified in fish and homeothermic vertebrates but not in poikilothermic vertebrates. In the present study, the expression of hsp90 alpha and hsp90 beta genes in Xenopus...
متن کاملOn the cellular and developmental lethality of a Xenopus nucleocytoplasmic hybrid
Nucleocytoplasmic hybrid (cybrid) embryos result from the combination of the nucleus of one species, and the egg cytoplasm of another species. Cybrid embryos can be obtained either in the haploid state by the cross-fertilization or intra-cytoplasmic injection of an enucleated egg with sperm from another species, or in the diploid state by the technique of interspecies somatic cell nuclear trans...
متن کاملIntegrin Linked Kinase (X-ILK) Function during Embryonic Development and within Adult Tissues of Xenopus laevis
Integrin linked kinase (ILK) is a serine/threonine protein kinase implicated in the phosphatidylinositol 3’kinase (PI3’K) pathway. Integrin linked kinase has been investigated in different organisms such as mammalian systems (human, mice, rat), insects (Drosophila) and nematodes (Cenorhabditis elegans), however to date little data regarding ILK research on amphibians has been reported. In...
متن کاملA description of the technique for nuclear transplantation in Xenopus laevis.
INTRODUCTION A METHOD by which nuclei can be successfully transplanted into Amphibian eggs was first worked out by Briggs & King (1952) for the eggs of Ranapipiens. Xenopus laevis is an atypical Anuran since its eggs can be obtained throughout the year, and the resulting embryos can be reared to maturity within 12 months under laboratory conditions. Because of these advantages we have used Xeno...
